Level and Effect of Adrenomedullin on Endotoxin Induced Disseminated Intravascular Coagulation Model in Rabbits

چکیده
Adrenomedullin (ADM) is a regulatory peptide having a variety of pharmacological properties secreted from vascular endothelial cells and adrenal medulla. There are only a few studies about the effects of ADM on coagulation and platelets which is secreted from endothelial cells. The aim of this study is to assay ADM levels in rabbits with endotoxin induced disseminated intravascular coagulation (DIC) model and to investigate the probable effects of ADM on coagulation parameters in this model. Four groups (Control, DIC, Heparin, ADM) each consisting of eight New Zeland rabbits were formed randomly. A DIC model was developed by infusion of endotoxin from Escherichia coli. Then the effect of standard dose heparin and 0.05μg/kg/min ADM infusion were evaluated. Administration of endotoxin resulted in severe changes of coagulation parameters as shown by the significant prolongation of the prothrombin time and activated partial thromboplastin time, decrease in platelet count, plasma fibrinogen level, antithrombin, and protein C activity. These results were in accordance with DIC findings and were significantly different from baseline and control group. In DIC group, the baseline, second, and sixth hour mean ADM levels were 7.09±1.63, 8.10±1.55, 9.35±0.76 ng/dl, respectively. ADM levels on sixth hour were significantly higher than baseline and control group. Heparin and ADM were used in the DIC model but both of them were not effective. In conclusion, ADM levels were found elevated during the progress of endotoxin induced DIC model. ADM were not effective in overt DIC model but it might be useful in the early phase of the DIC.

Immunodepletion of extrinsic pathway inhibitor sensitizes rabbits to endotoxin-induced intravascular coagulation and the generalized Shwartzman reaction.
We have reported earlier that immunodepletion of extrinsic pathway inhibitor (EPI) sensitizes rabbits to disseminated intravascular coagulation (DIC) induced by infusing a low concentration of tissue factor (TF). We now describe the effect of immunodepletion of EPI in rabbits administered endotoxin. Cortisone-treated rabbits were administered anti-rabbit EPI immunoglobulin (IgG) or Fab fragment...

We evaluated the effect of r-hirudin on an experimental model of disseminated intravascular coagulation (DIC) in rabbits, through the continuous infusion of 100 microg/kg/hr of Escherichia coli endotoxin for a period of 6 hours. r-Hirudin (0.05, 0.3, and 0.6 mg/kg/hr) as treatment, or saline solution as placebo, were administered simultaneously with endotoxin. Severe DIC in the endotoxin contro...

The effect of low molecular weight heparin (LMWH) with or without antithrombin III (AT III) has been studied in a rabbit model of disseminated intravascular coagulation (DIC) induced by continuous infusion of 100 microg/kg/hr of Escherichia coli endotoxin for 6 hr. LMWH (5 and 10 IU/kg/hr/6 hr), alone or in combination with AT III (20 U/kg/hr/6 hr), or saline were administered simultaneously wi...
متن کاملThe role of granulocytes in the activation of intravascular coagulation and the precipitation of soluble fibrin by endotoxin.
This study examines the role of neutrophils (PMN) in the pathogenesis of endotoxin-induced microclot formation. It is intended to clarify whether granulocytes are involved in endotoxin-induced activation of intravascular coagulation (generation of soluble fibrin) and/or in endotoxin-induced precipitation of soluble fibrin. Precipitation of soluble fibrin was achieved by injection of endotoxin i...
متن کاملDisseminated intravascular coagulation in rabbits induced by administration of endotoxin or tissue factor: effect of anti-tissue factor antibodies and measurement of plasma extrinsic pathway inhibitor activity.
Rabbits were given polyclonal anti-tissue factor (TF) immunoglobulin G (IgG) before an injection of endotoxin to test the hypothesis that TF triggers disseminated intravascular coagulation (DIC) after endotoxin. The rabbits had been prepared with cortisone to develop DIC after one injection of endotoxin. Anti-TF IgG substantially reduced the falls in fibrinogen, factors V and VIII, and platelet...
